A chromEM-staining protocol optimized for cardiac tissue

چکیده
Three-dimensional (3D) chromatin organization has a key role in defining the transcription program of cells during development. Its alteration is cause gene expression changes responsible for several diseases. Thus, we need new tools to study this aspect regulation. To end, ChromEM was recently developed: an electron-microscopy staining technique that selectively marks nuclear DNA without altering its structure and, thus, allows better visualization 3D conformation. However, despite increasingly frequent application on cells, it not yet been applied visualize ultrastructure tissues. Here, provide protocol carry out myocardial tissue harvested from left ventricles C57BL/6J mice and use combination with transmission electron microscopy (TEM) measure some morphological parameters peripheral heterochromatin cardiomyocytes. This could also be used, tomography, cardiomyocytes different aspects heart pathobiology (e.g., development, cardiac aging, failure) as well help set-up other

A Protocol for Collecting Human Cardiac Tissue for Research
This manuscript describes a protocol at the University of Kentucky that allows a translational research team to collect human myocardium that can be used for biological research. We have gained a great deal of practical experience since we started this protocol in 2008, and we hope that other groups might be able to learn from our endeavors. To date, we have procured ~4000 samples from ~230 pat...
متن کاملOptimized Protocol of Methanol Treatment for Immunofluorescent Staining in Fixed Brain Slices
We optimized methanol treatment in paraformaldehyde-fixed slices for immunofluorescent staining of ependymal basal bodies in brain ventricles. As 100% methanol induced severe deformations to the slices (including rolling and folding over), we tried to decrease methanol concentration. We found that 33.3% to 75% methanol could result in ideal immunostaining of basal bodies without inducing obviou...
متن کاملAn optimized negative-staining protocol of electron microscopy for apoE4•POPC lipoprotein
Apolipoprotein E (apoE), one of the major protein components of lipoproteins in the peripheral and central nervous systems, regulates cholesterol metabolism through its interaction with members of the low density lipoprotein receptor family. One key to understanding apoE function is determining the structure of lipid-bound forms of apoE. Negative-staining (NS) electron microscopy (EM) is an eas...
متن کاملA robust RNA integrity-preserving staining protocol for laser capture microdissection of endometrial cancer tissue.
Laser capture microdissection of frozen tissue sections allows homogeneous cell populations to be isolated for expression profiling. However, this requires striking a balance between retaining adequate morphology for accurate microdissection and maintaining RNA integrity. Various staining protocols were applied to frozen endometrial carcinoma tissue sections. Although alcohol-based methods were...
متن کاملOptimized Negative Staining: a High-throughput Protocol for Examining Small and Asymmetric Protein Structure by Electron Microscopy
Structural determination of proteins is rather challenging for proteins with molecular masses between 40 - 200 kDa. Considering that more than half of natural proteins have a molecular mass between 40 - 200 kDa, a robust and high-throughput method with a nanometer resolution capability is needed. Negative staining (NS) electron microscopy (EM) is an easy, rapid, and qualitative approach which h...
